Anti-oxidant and Anti-obese Effects of Mulberry (Morus alba L.) Leaf Extract in 3T3-L1 Cells

Abstract
Recently, numerous studies reported that excessive generation of ROS stimulated expression of adipogenesis transcription factors, such as C/EBP¥á, ¥â, and PPAR¥ã in 3T3-L1 cells. Therefore, effective scavenging of ROS could be regarded as strategy for anti-adipogenic effect through suppression of adipogenesis transcription factors. In this study, we examined the anti-oxidant and anti-adipogenic effects of mulberry (Morus alba L.) leaf extract (MLE). MLE was prepared by boiling water extraction method at 121¡ÆC for 15 min. DPPH radical scavenging activity of MLE was significantly increased to 3.48%, 8.28%, 16.63%, and 25.84% between 100 and 1,000 ¥ìg/mL. The MLE treatment for 24 h did not affect to the cell viability of confluent 3T3-L1 preadipocytes at concnetrations of 10, 50, 100, 200, 400, and 800 ¥ìg/mL. Thus, nontoxic concentration rages of MLE were treated during adipogenesis period (Day -2 to 6) from preadipocytes into adipocytes in 3T3-L1 cells. Intracellular lipid accumulation in MLE-treated 3T3-L1 adipocytes (Day 6) were quantitatively judged by ORO staining and a number of lipid droplet and size were observed using microscopy. Inhibitory activity of lipid accumulation by MLE treatment in 3T3-L1 adipocytes was significantly increased 19.52%, 16.98%, and 32.48% at 200, 400, and 800 ¥ìg/mL, respectively. The size and a number of lipid droplet in 3T3-L1 adipocytes were observed to dose-dependently decrease by MLE treatment. The mRNA expression levels of C/EBP¥á, ¥â and PPAR¥ã were decreased at 10 and 100 ¥ìg/mL in MLE-treated 3T3-L1 adipocytes compared with adipocytes. These results showed that antioxidant activity of MLE was contributed to anti-obesity effect by inhibition of intracellular lipid accumulation through suppression of adipogenesis transcription factors, such as C/EBP¥á, ¥â, and PPAR¥ã. Therefore, MLE is expected to utilize as anti-oxidant and anti-obesity cosmetic material.
